Structure-switching aptamers
WebNov 5, 2024 · We identified structure-switching aptamers (SSAs) that specifically bind to HSA with low nanomolar dissociation constant via three rounds of MCP-SELEX and 1 round of falling-off-SELEX. Our HSA SSAs also have ∼3-fold higher specificity against streptavidin relative to thrombin SSAs discovered through falling-off-SELEX only. WebOct 4, 2024 · Aptamers can form several types of secondary structure; one example is shown in part b — the predicted secondary structure of a DNA aptamer binding to human α-thrombin.

Nucleic acid aptamers are short single-stranded DNA or RNA developed by SELEX (systematic evolution of ligands by exponential enrichment) process [1].Unlike antibodies, which act primarily as antigenic epitopes, aptamers assumed unique three-dimensional structures and can selectively interact with targets by non-covalent …
WebOct 15, 2024 · Structure-switching aptamers (SSAs) offer a promising recognition element for sensor development. However, the generation of SSAs via in vitro aptamer selection technologies or post-selection ... WebAptamers are widely employed as recognition elements in small molecule biosensors due to their ability to recognize small molecule targets with high affinity and selectivity. Structure-switching aptamers are particularly promising for biosensing applications because target-induced conformational change can be directly linked to a functional output.
